What's the context of your question? Where is the billboard? What's the traffic volume? Who are you trying to attract/repulse? What's the action you want viewers to take? What images/messages have you tried to date (in other media formats, for example)?

This billboard will be for the youth not to aware them about the affect of harmful drugs.

You can check out some anti-drug billboards used by looking doing a Google image search on "anti-drug billboards". Here is a,link to it: https://tinyurl.com/myb3ep8.

The country/culture makes a big difference on what would work, so you might want to specify that in the search

Most billboards are a waste of money and a blight on the landscape. Instead, focus your efforts on group advocacy sessions in which you have young people who have used drugs and who have come out the other side tell their stories.

Whatever you do about your billboard, test your thoughts on the social media sites - Instrgram, Twitter et al. That way you'll get it right before you hit the road, as it were.

So what can you say? After all, most drug use is the result of some kind of dissociation from family or society. In other words, the problem you face is that drugs are the symptom not the cause. I speak as one who meditates consistently and as a youngster at uni was astonished to hear a friend describe my "trip" in some detail - but he'd been on LSD. Just to give you an idea ...

Using this approach you could say "What are you missing out on?"
"What changed that you needed a fix instead?"
What changed that meant you needed a fix just to cope?
Can you cope without a fix?
"Who talked you into taking a hit?"

Because the tired line "don't take drugs" doesn't work, and never did. You have to be pro-active. Just as a side-issue here in NL, the rate of drug addiction is well below that of Anglo-Saxon countries. And it's legal. Think about it!!

I'll add that from a Dutch study, Steiner-Waldorf educated kids are around 10x less likely to become drug addicts. There are sound reasons as to why this should be so, and it's all to do with their emotional security.
