Need to begin following people and building up your followers. On your home page use the 'find people' tool at the top to search for people with similar interests. You can type in by name or topic and it will display the usernames that are relevant. There are many tools that can be used in conjunction with Twitter to get the most out of your experience. I would recommend getting the book "Twitter Revolution: How Social Media and Mobile Marketing is Changing the Way We Do Business & Market Online" by by Warren Whitlock and Deborah Micek. It has a lot of great tips to get you started.
